آفت‌کش‌های کشاورزی از ساختار پیچیده مولکولی برخوردارند و به عنوان ابزار قدرتمندی برای تأمین نیازهای غذایی جمعیت در حال رشد جهان، توسعه و کاربرد فراوانی یافته‌اند. مصرف بی‌رویه این مواد را تبدیل به تهدیدی جدی برای محیط‌زیست کرده است. همه ساله مقدار زیادی از آفت‌کش‌ها از طرق مختلف به محیط زیست راه پیدا می‌کنند. این آفت‌کش‌ها با تأثیر بر شرایط خاک، تخریب و کاهش توان زراعی، اثرات مخربی بر کیفیت زندگی انسان می‌گذارند. آفت‌کش‌های فسفره آلی دسته ای از نوروتوکسین‌های بسیار سمی هستند و به علت مسمومیت بالایی که برای حشرات، پستانداران و سایر حیوانات دارند، این ترکیبات، مهارکننده آنزیم استیل کولین استراز و مانع انتقال پیام‌های عصبی هستند. دیازینون یکی از آفت‌کش‌های فسفره آلی است که در کنترل طیف وسیعی از آفات استفاده می‌شود. بررسی‌های زیادی در مورد اثرات سمیت این آفت‌کش در محیط زیست انجام گرفته است. برای حذف این آلودگی‌ها از محیط زیست روش ‌های مختلفی استفاده می‌شود. اما استفاده از میکروارگانیسم‌ها از جمله باکتری‌ها برای پاکسازی این آلودگی‌ها موثرتر و ارزان‌تر می‌باشد. این پژوهش با هدف جداسازی باکتری‌های بومی تجزیه‌کننده دیازینون از مناطق آلوده صنعتی انجام شد. جداسازی باکتری‌های تجزیه‌کننده این آفت‌کش با کشت نمونه‌ها روی محیط‌کشت معدنی صورت گرفت. ارزیابی قدرت تجزیه‌کنندگی باکتری‌ها با استفاده از روش کروماتوگرافی مایع با کارایی بالا انجام شد. در نهایت شناسایی مولکولی سویه‌های برتر با استفاده از روش 16s RDNA انجام گرفت. از بین باکتری-های جداسازی شده باکتری Pseudomonas stutzeri بیشترین توانایی را در تجزیه دیازینون نشان داد. این جدایه غلظت آفت‌کش را طی 15 روز از غلظت اولیه‌ی50 میلی گرم بر لیتر به 15میلی‌گرم بر لیتر رساند. با توجه به رشد باکتری‌ها در محیط‌کشت‌های فقیر چنین استنباط می‌شودکه این جدایه‌ها از این دو ترکیب به عنوان منبع انرژی برای رشد خود استفاده می‌کنند. بنابراین می‌توان از این جدایه‌ها پس از بررسی در سطح میدان و تعیین فرمولاسیون آن‌ها، جهت پاکسازی زیستی استفاده نمود.

Agricultural pesticides are complex molecular structures which are highly developed as a means of fulfilling nutritional needs of the growing world population. Each year, a significant amount of pesticides leaks into the environment through different ways. Pesticides are distributed throughout the environment and move further into remote areas. They affect soil conditions, destruct and reduce the crop and therefore, have damaging effects on the quality of human life. Organic phosphorus pesticides are a group of highly toxic neurotoxin and this poisoning effect on insects, mammals and other animals causes them to inhibit the acetylcholinesterase enzyme and prevent the transmission of information by neurons. Diazinon is one of the organic phosphorus pesticides used to control a wide range of pests. A lot of research has been done on the effects of this pesticide on the environment. Different methods are used to remove these contaminants from the environment. But the use of microorganisms, including bacteria, is more effective and cheaper to clean up these contaminants. This research was carried out with the aim of isolating indigenous bacteria from diazinon degradation from industrial contaminated areas. Isolation of the pesticide degrading bacteria by culture of the samples was performed on the mineral medium. Ultimately, molecular identification of superior strains was performed using the RDN 16s method. Among the isolated bacteria, Pseudomonas stutzeri showed the greatest ability to decompose diazinon. The isolate produced a concentration of pesticide within 15 days of initial concentration of 50 mg. L to 15 mg. L. Given the growth of bacteria in poorer environments, it is thought that these isolates use these two compounds as sources of energy for their growth. Therefore, these isolates can be used for biological purification after examining the field surface and determining their formulation.
