گزارش پوسیدگی نرم سیر ناشی از Pectobacterium carotovorum در استان لرستان

چکیده
سیر از محصولات مهم زراعی در ایران و استان لرستان به شمار می‌آید. از بیماری‌های مهم باکتریایی سیر که باعث کاهش تولید آن می‌شود پوسیدگی ‌نرم سیر است که طی چند سال اخیر، تولید این محصول را در استان با تهدید روبرو کرده است. در انجام این تحقیق، نمونه‌هایی با علائم لهیدگی از مزارع سیر استان جمع‌آوری و به آزمایشگاه منتقل گردید. پس از ضدعفونی سطحی با محلول هیپوکلریت پنج درصد، قطعاتی از بافت دارای علائم در مقداری آب مقطر سترون در هاون‌های سترون له گردید و پس از گذشت دو تا سه ساعت چند قطره از عصاره مذکور در محیط‌های کشت نوترینت آگار مغذی (NA) و (EMB)ایوزین متیل بلو وکینگ-ب (king B) کشت شد و در انکوباتور با دمای 28 درجه سانتی‌گراد به مدت سه الی چهار روز نگهداری شد. پس از ظهور کلنی‌ها بر روی محیط کشت، کلنی‌هایی هدف انتخاب و به‌منظور تهیه کشت خالص مجدد کشت گردیدند. جهت انجام آزمون بیماری‌زایی بر روی سیر، سوسپانسیونی با غلظت 108 واحد تشکیل‌دهنده کلنی در میلی‌لیتر در داخل زخم‌های ایجادشده در بوته سیر تلقیح شد. جهت تائید آزمون بیماری‌زایی، پس از گذشت یک هفته علائم مذکور مشاهده و مجدداً خالص‌سازی گردید. آزمون‌های فیزیولوژیک و بیوشیمیایی بر اساس روش‌های استاندارد باکتری‌شناسی تا حد شناسایی گونه انجام شد. بر اساس نتایج آزمون‌های بیوشیمیایی و همچنین استفاده از آغازگرهای PCCSSF/PCCSSR و تکثیر ژن‌های کد کننده ناحیه16S rRNA با استفاده از آغازگرهای FD1/ rD1در آزمون PCR، جدایه-های مذکور به‌عنوان Pectobacterium carotovorum شناسایی شدند. این اولین گزارش از وقوع این عامل بیماری در استان لرستان است

First report of garlic soft rot caused by Pectobacterium carotovorum in Lorestan province

Abstract
Garlic is considered as one of the most important crops in Iran and specially in Lorestan Province. Soft rot is one of the important bacterial diseases of garlic, which reduces yield, and has threatened garlic production in Lorestan province in recent years. In this study, samples showing symptom of soft rot were collected from garlic fields and transferred to the laboratory. After surface sterilization with 5% sodium hypochlorite solution, tissues with symptoms were squashed in the mortar containing sterilized distilled water under sterile condition. Subsequently, a few drops of extract were cultured on nutrient agar (NA), Eosin methylene blue (EMB) and King's B and incubated at 28 °C for three or four days. After the emergence of colonies, target colonies were selected based on colony morphology and shape. To obtain purified colonies, colonies were sub-cultured on nutrient agar. To carry out pathogenicity test, a bacterial suspension (5 µl) containing 108 colony-forming units per ml (CFU ml-1) was inoculated into the wounds. To confirm Koch’s postulates, bacteria re-isolated from diseased bulbs were streaked on NA and reidentified. Physiological and biochemical features were characterized based on standard bacteriological criteria to the species level. A pair of specific primers (PCCSSF / PCCSSR) was used to detect these bacterial strains. Based on the results of biochemical tests and molecular identification using PCCSSF / PCCSSR and 16s rRNA amplifying primers (FD1 / rD1), the isolates were identified as Pectobacterium carotovorum. To the best of the knowledge, this is the first report of the occurrence of this disease in Lorestan province.
