بررسی احتمال همراهی پروکاریوت‌های محدود به آوند آبکشی در درختان نارنگی سیاهو با علایم بدشکلی و تغییر رنگ غیرطبیعی میوه

چکیده
در سال‌های اخیر در برخی از درختان نارنگی در منطقه‌ی سیاهو علایم بدشکلی، عدم تقارن و تغییر رنگ غیرطبیعی در میوه‌ها مشاهده شده که منجر به عدم بازارپسندی و حتی غیرقابل برداشت شدن محصول توسط باغدار شده است. علایم ایجاد شده در این درختان، به همراه علایمی مانند کمبودهای مواد غذایی، پیسک و زردی که در برگ‌های برخی از این درختان مشاهده می‌شوند، شبیه به علایم بیماری‌هایی مانند هوانگ‌لونگ‌بینگ مرکبات، استابورن مرکبات و همچنین برخی از بیماری‌های فیتوپلاسمایی می‌باشد. به منظور بررسی‌ نقش پروکاریوت‌های محدود به آوند آبکشی (Candidatus Liberibacter spp.، Spiroplasma citri و Ca. Phytoplasma spp.) با علایم تغییر رنگ غیرطبیعی و بدشکلی میوه‌های نارنگی در منطقه‌ی سیاهو، 40 درخت دارای علایم در میوه و پنج درخت بدون علایم شناسایی و شماره‌گذاری شد و نمونه برداری به طور عمده از دم‌میوه‌ی میوه‌های دارای علایم این درختان انجام شد. ردیابی مولکولی این پروکاریوت‌ها با آزمون PCR با استفاده از آغازگرهای اختصاصی این بیمارگرها در نمونه‌هایDNA استخراج شده از دم‌میوه‌ها نشان داد که در همه‌ی درختانی که علایم تغییر رنگ غیرطبیعی به همراه بدشکلی میوه وجود داشت باکتری Candidatus Liberibacter asiaticus، عامل فرم آسیایی بیماری هوانگ‌لونگ‌بینگ مرکبات، ردیابی شد. در هیچ درخت دارای علایمی وجود فیتوپلاسما اثبات نشد و تنها در چهار درخت، Spiroplasma citri، عامل استابورن مرکبات، به همراه Ca. L. asiaticus ردیابی شد. در پنج درخت بدون علایم، پروکاریوت‌های نامبرده ردیابی نشدند. برای بررسی صحت ردیابی‌ها، تعیین ترادف برخی از قطعات تکثیر شده در PCR و جستجو با برنامه-ی بلاست با استفاده از این ترادف‌ها در بانک جهانی ترادف‌ها انجام و گونه‌ی Ca. L. asiaticus تایید شد. نتایج به دست آمده از میزان ردیابی این پروکاریوت‌ها در درختان دارای علایم نشان داد که باکتری Ca. L. asiaticus با علایم تغییر رنگ غیرطبیعی و بدشکلی میوه در درختان نارنگی سیاهو همراه می‌باشد و این نشان دهنده‌ی وقوع و گسترش بیماری هوانگ‌لونگ‌بینگ مرکبات در این منطقه است.

Investigation of association of phloem-limited prokaryotes with Siyahoo tangerine trees showing misshapen and inverted color change symptoms in fruit

Abstract
In recent years, symptoms of abnormal color changes and deformity have been observed in fruits of several tangerine (Citrus reticulata cv. Siyahoo) trees in Siyahoo region (Hormozgan province), which has led to lack of marketability and economic losses. These symptoms on fruits along with the symptoms like nutrient deficiencies, mottling and yellowing in leaves were similar to the symptoms caused by huanglongbing (HLB), stubborn and phytoplasma diseases. To investigate the association of phloem-limited prokaryotes (Ca. Phytoplasma spp., Ca. Liberibacter spp. and Spiroplasma citri) with Siyahoo tangerine trees showing misshapen and inverted color change symptoms in fruit, 40 symptomatic and five symptomless trees were identified and numbered. Sampling was mainly done from peduncles of symptomatic fruits and DNA was extracted from them. Molecular detection of these prokaryotes by PCR assay using specific primers revealed that Candidatus Liberibacter asiaticus, the causal agent of Asian HLB, was detected in all trees showing fruit color inversion. There was no evidence of phytoplasma presence in any of the symptomatic trees. Spiroplasma citri, the causal agent of citrus stubborn disease, was detected in only four trees with symptomatic fruits and leaves. None of these prokaryotes were detected in five symptomless trees. Sequencing of some PCR products, and BLAST search using these sequences in GenBank database confirmed the presence of Ca. L. asiaticus in symptomatic trees. In conclusion, PCR detection rate of these prokaryotes in suspected trees revealed that Ca. L. asiaticus is associated with misshapen and inverted color change in tangerine fruit. This result indicates the occurrence and spread of HLB disease in tangerine orchards in Siyahoo region.
