اثر بازدارندگی باکتری CHA0 Pseudomonas fluorescensو عصاره‌ی آبی برخی گیاهان بر نماتد ریشه‌گرهی (Meloidogyne javanica) در گیاه گوجه‌فرنگی

چکیده
نماتدهای ریشه‌گرهی (Meloidogyne spp.) یکی از مهم‌ترین عوامل محدود کننده‏ی رشد گیاهان هستند که گسترش جهانی و تعداد زیادی میزبان دارند. باکتری Pseudomonas fluorescens CHA0 (Pf) توانایی مهار نماتدهای ریشه‌گرهی را دارد. اخیراً تمایل به استفاده از نماتدکش‌های دوست‌دار محیط زیست مانند عصاره‌های گیاهی به جای سموم شیمیایی و همچنین عوامل بیوکنترل افزایش یافته است. در مطالعه حاضر، تأثیر یک ایزوله از باکتری Pf و دو عصاره گیاهی شامل مورد (Myrtus cominus) و تاتوره (Datura stramonium) بر نرخ تفریخ تخم و مرگ ‌و میر لارو سن دوم نماتد M. javanica در شرایط آزمایشگاه مورد ارزیابی قرار گرفت. بعد از تعیین غلظت کشنده 30، 50 و 70 درصد (LC30، LC50 و LC70) مربوط به هر کدام از عصاره‌های گیاهی، بذر گوجه‌فرنگی رقم Early-Urbana در گلدان‌های پلاستیکی دو کیلوگرمی حاوی بستر کشت استریل کشت گردید. بستر کشت گیاهچه‌ها در مرحله چهار برگی با 20 میلی‌لیتر سوسپانسیون 108 CFU/ml از باکتری Pf تیمار شدند. یک هفته بعد، گیاهان به طور هم‌زمان با 4000 تخم و لارو سن دوم نماتد و 100 میلی‌لیتر از غلظت‌های انتخاب شده از گیاه تاتوره شامل 1/1، 4/1 و 8/1 درصد وزن به حجم و غلظت‌های 8/1، 3 و 2/5 درصد وزن به حجم از گیاه مورد تیمار شدند. مطالعات آزمایشگاهی در قالب طرح کاملاً تصادفی و مطالعات گلخانه به صورت فاکتوریل در قالب طرح کاملاً تصادفی با چهار تکرار انجام شدند. شاخص‌های رشدی گوجه‌فرنگی همچنین شاخص‌های جمعیتی نماتد پس از 60 روز مورد ارزیابی قرار گرفتند. نتایج نشان داد که ترکیب باکتری Pf و عصاره‌های گیاهی باعث بهبود شاخص‌های رشدی گیاه و کاهش شاخص‌های جمعیتی نماتد می‌گردد. استفاده از باکتری Pf در ترکیب با غلظت 8/1 درصد وزن به حجم عصاره‏ی تاتوره و 2/5 درصد وزن به حجم عصاره‏ی مورد در مقایسه با شاهد به عنوان موثرترین تیمار بودند که به ترتیب باعث کاهش تعداد تخم در ریشه و همچنین فاکتور تولیدمثل نماتد به میزان 68 و 45 درصد، تعداد گال در ریشه به میزان 64 و 33 درصد و تعداد توده تخم در ریشه به میزان 65 و 43 درصد گردید.

Abstract
Root-knot nematodes which have worldwide distribution and lots of host plants are one of the major limiting factors affecting plant growth. Pseudomonas fluorescens CHA0 (Pf) have the potential for controlling the root-knot nematodes. There is an interest to use eco-friendly nematicides such as different plant extracts, instead of chemical pesticides. In the present study, an isolate of Pf and different concentrations of two plants extracts, namely Datura stramonium and Myrtus cominus were investigated on egg hatching and larval mortality of M. javanica under laboratory conditions. After determination of the LC30, LC50 and LC70 values of each extract, seeds of tomato cv. Early-Urbana were planted in plastic pots containing 2kg of steam-sterilized soil mixture. Four leaf stage seedlings of tomato were treated with 20 ml of Pf suspension at a concentration of 108 CFU/ml, as soil drench. One week after treatment, they were inoculated with 4000 eggs and second stage juveniles of M. javanica and simultaneously were treated with 100 ml of selected concentrations viz. 1.1, 1.4 and 1.8% (W/V) of D. stramonium and also selected concentrations viz. 1.8, 3 and 5.2% (W/V) of M. cominus were added to the soil. The in vitro experiment was conducted as a completely randomized design (CRD) and in vivo experiment as a factorial in completely randomized design with four replicates. Plant growth as well as nematode population indices were evaluated 60 days after inoculation. Results showed that plant extracts in combination of Pf increased the growth of plants and decreased the nematode indices. The treatment with combination of Pf and leaf extract of D. stramonium at the rate of 1.8% or M. cominus at the rate of 5.2%, was the most effective treatment. As compared to control, the reduction in number of eggs per root system as well as the reproductive factor, respectively was 68 and 45%, reduction in number of galls per root system, respectively was 64 and 33% and reduction in number of egg masses, respectively was 65 and 43%.
