چکیده
در این آزمایش، القای مقاومت گوجه‌فرنگی (رقم سوپرچیف) به ویروس موزاییک خیار (CMV) از طریق کاربرد اسید سالیسیلیک (SA) و جاسمونیک (JA) مورد ارزیابی قرار گرفت. تیمارها شامل CMV، SA (1 میلی‌مولار)، JA (5/0 میلی‌مولار)، SA+JA، SA+CMV، JA+CMV، SA+JA+CMV و شاهد بود. CMV به صورت مکانیکی با استفاده از کربوراندوم روی گوجه‌فرنگی مایه‌زنی شد. نمونه‌ها در صفر، 1، 2، 4، 6، 8 و 15 روز پس از مایه‌زنی تهیه شدند. بر اساس نتایج، شدت فتوسنتز و میزان کلروفیل a و b در تیمارهای حاوی ویروس و هورمون کاهش یافت. SA موجب افزایش میزان کلروفیل a و b طی زمان در مقایسه با شاهد گردید، در حالی که JA فقط میزان کلروفیل a را افزایش داد و اثر منفی بر مقدار کلروفیل b داشت. بیان ژن پوشش پروتئینی CMV (CMV-CP) در تیمار SA+JA+CMV به میزان 48 درصد نسبت به تیمار مایه‌زنی با CMV در روز پانزدهم کاهش داشت. در تیمار شاهد و سایر تیمارهای هورمونی فاقد CMV، ژن CMV-CP قابل ردیابی ملاحظه نگردید. بیشترین میزان بیان ژن‌های PR1 و PR10 در تیمار SA+JA+CMV مشاهده شد که به ترتیب در حدود 28 و 24 درصد بیشتر از بیان این ژن‌ها در تیمار CMV طی روزهای ششم و هشتم نمونه‌برداری بود. بیان PR1 در تیمارهای حاوی CMV بیشتر از تیمارهای هورمونی فاقد ویروس بود. بیشترین میزان بیان PR10 در تیمار SA+JA+CMV و روز هشتم مشاهده شد و تا روز پانزدهم کاهش یافت. در همه تیمارها، وزن خشک گوجه‌فرنگی در مقایسه با شاهد کاهش نشان داد. بیشترین میزان کاهش وزن خشک (47 درصد) در تیمار JA+CMV دیده شد که با تیمار CMV (44 درصد) تفاوت معنی‌داری نداشت. در مجموع، ترکیب دو هورمون SA و JA، مقاومت گوجه‌فرنگی (رقم سوپرچیف) به CMV را بهبود بخشید و وزن خشک اندام هوایی گیاه را به میزان 22 درصد در مقایسه با تیمار CMV افزایش داد.

Abstract
Induction of resistance to Cucumber mosaic virus (CMV) in tomato plants (cv. Superchief) by application of salicylic acid (SA) and jasmonic acid (JA) was evaluated in this experiment. Treatments included CMV, SA (1 mM), JA (0.5 mM), SA+JA, SA+CMV, JA+CMV, SA+JA+CMV and a control. CMV was mechanically inoculated on tomato plants using carborundum. Samples were taken at 0, 1, 2, 4, 6, 8, and 15 days post inoculation (dpi). Based on the results, photosynthesis rate and chlorophyll a and b content decreased under the virus infection and hormone treatment. SA increased both chlorophyll a and b contents in comparison to the control over the time, but JA only increased the chlorophyll a content and had a negative effect on chlorophyll b content. Expression of CMV coat protein gene (CMV-CP) was decreased approximately 48% in SA+JA+CMV treatment in comparison to the CMV- infected plants at 15 dpi. The control and hormone-treated without CMV plants had no detectable CMV-CP gene. The highest expression rate of PR1 and PR10 observed in SA+JA+CMV treatment that were 28 and 24% greater than CMV at 6 and 8 dpi, respectively. Expression of PR1 was greater in treatments consisting CMV in comparison to hormone treatments without CMV. Expression of PR10 was the highest in SA+JA+CMV at 8 dpi and reduced at 15 dpi. Tomato plants dry weight decreased in comparison to control in all treatments. The highest dry weight loss (47%) observed in JA+CMV that was not significantly different with CMV (44%). In conclusion, combination of SA and JA improved the resistance against CMV in tomato (cv. Superchief) and increased aerial dry weight about 22% compared with CMV control.
