چکیده
بررسی به منظور جداسازی و شناسایی نماتودهای بیمارگر حشرات از سه منظقه‌ی مهم کشور عراق ، انجام شد. نمونه‌های خاک از پانزده محل شامل علفزارها، باغات خرما و مرکبات از این سه استان جمع‌آوری و حضور احتمالی نماتودهای بیمارگر حشرات در این نمونه‌ها مورد بررسی قرار گرفت. از لارو سن آخر پروانه‌ی موم‌خوار بزرگ(Lep.: Pyralidae) Galleria mellonella، به‌عنوان تله برای جداسازی نماتودهای بیمارگر احتمالی استفاده گردید. پس از سه تا چهار روز، اجساد لاروهای مرده (مشکوک به آلودگی به نماتود) به تله وایت منتقل شد تا لاروهای عفونت‌زای نماتود پس از خارج شدن از لاشه‌ی گالریا جمع‌آوری گردند. سپس، لاروهای عفونت‌زای جمع‌آوری شده در لوله‌های فالکون به‌همراه مقدار اندکی آب در دمای هفت درجه‌ی سانتیگراد نگهداری شدند. پس از استحصال نماتود از آنها، سویه های جدا شده با استفاده از داده های مرفومتریک و مولکولی شناسایی شد.در بین نمونه های جمع اوری شده جدایه ای منسوب به جنس Heterorhabditis بود.صفات مرفولوژیک و مرفومتریک کلیدی افراد بالغ نر، ماده و لارو عفونت زا نشان داد که جدایه مذکور به گروه گونه ای bacteriophora تعلق دارد. تکثیر و توالی یابی نواحی ژنی ITS و 18Sو سپس تحلیل شجره شناسی مبتنی بر آن، محرز ساخت که جدایه مذکور در کنار جدایه های دیگری از گونه H.bacteriophora در یک شاخه قرار گرفت. میزان فواصل نوکلئوتیدی نیزاین تایید را نشان داد.این گونه اولین گزارش نماتود بیمارگر حشرات از عراق است. این ایزوله با ناحیه های ITS و 18S در عراق شناسایی شده وتوالی این ایزوله نشان داده که بیشترین شباهت را باکستان وایران دارد واین می تواند به خاطر نزدیک قرابت جغرافیایی دانس است

Heterorhabditis bacteriophora, a pioneer in Iraq for entomopathogenic nematode

Abstract
A survey was conducted in three main regions of Iraq to isolate and identify entomopathogenic nematodes (EPN). Soil samples were collected from 15 sites across the tree provinces of grassland, date palm and citrus fields and tested to present species of EPN. Last stage of Galleria mellonella (Lep.: Pyralidae) were used as a trap to collect and isolate the EPNs. After 3-5 days, the infected larvae were transfer to white trap to be able to collect the invective juveniles (IJs) after emergence. After that, the IJs that were collected, stored in shallow water into falcon tube at 7 °C. All nematode specimens used in this study were reared in G. mellonella larvae. From a total of 75 samples, 50% of samples tested positive for the present of nematodes. The collected specimens were characterized first morphologically, then molecular studies were conducted to characterize and identify the isolates on the bases of internal transcribed spacer (ITS) and 18S rDNA regions. The collected samples were from Heterorhabditis genus. The phylogenetic relations of Heterorhabditis species were analyzed using the maximum likelihood method. The BLAST analysis of the ITS and 18S regions of the rDNA of the Heterorhabditis isolate IRQ showed a rather high similarity of 99% with Heterorhabditis bacteriophora (MF033536). The identified species was as isolate of Heterorhabditis as Heterorhabditis bacteriophora, this is the first date of ITS rDNA and 18S H. bacteriophora from Iraq (Baghdad province). Analysis of Iraqi H. bacteriophora showed that Iraqi isolate place close to Iranian and Pakistan isolates. This could be because the similar environment among these places.
