شناسایی قارچ‌های اندوفیت گیاهان Glaucium fimbrilligerum، Chelidonium majusو Papaver macrostomum از تیره خشخاش در استان گلستان

چکیده
قارچ‌های اندوفیت‌ از عواملی هستند که حداقل قسمتی از چرخه زندگی خود را درون بافت گیاهی بدون ایجاد علائم قابل مشاهده‌ای، زندگی می‌کنند. تحقیقات نشان داده است که نزدیک به 300000 گونه گیاهی در زمین وجود دارد و هر گیاه می‌تواند میزبان یک و یا تعداد بیشتری قارچ اندوفیت باشد. این قارچ‌ها منبع اصلی ترکیبات جدید مورد استفاده در پزشکی، کشاورزی و صنعت هستند. به منظور شناسایی قارچ‌های اندوفیت گیاهان Glaucium fimbrilligerum، Chelidonium majusو Papaver macrostomum از تیره‌ی خشخاش، طی بهار و تابستان سال 1396 در استان گلستان نمونه‌برداری از محل گره بوته‌های سالم انجام شد. ضدعفونی سطحی نمونه‌ها به ترتیب با استفاده از اتانول ٪70 به مدت 45 ثانیه، هیپوکلریت سدیم یک درصد به مدت 30 ثانیه و سپس سه بار شستشو با آب مقطر سترون انجام گرفت. نمونه‌ها در تشتک‌های پتری حاوی محیط کشت سیب‌زمینی- دکستروز- آگار کشت و به مدت 4 هفته در انکوباتور با دمای C°25 نگه‌د‌اری شدند. پرگنه‌های رشد یافته از حاشیه‌ی نمونه‌ها، جداسازی و به روش تک اسپور و با نوک ریسه خالص‌سازی شدند. پس از خالص‌سازی هر یک از جدایه‌ها بر اساس خصوصیات مورفولوژیک مانند اندازه کنیدی و کنیدیوفور و با استفاده از کلیدهای شناسایی معتبر شناسایی شدند. برای شناسایی جدایه‌های بدون اسپور و بررسی صحت شناسایی‌های انجام شده بر اساس ویژگی‌های مورفولوژیک، شناسایی مولکولی و توالی نواحی ITS (ITS4 و ITS5) ازDNA ریبوزومی انجام و سپس با بانک ژن مقایسه شد. در این بررسی سه گونه‌ی قارچی Alternaria japonica، Alternaria arborescens و Lopadostoma fagi به عنوان قارچ‌های اندوفیت از گیاهان مورد تحقیق شناسایی شدند. گونه‌های آلترناریا و یک جنس جدید برای اولین بار از روی گیاهان تیره‌ی خشخاش گزارش می‌شوند.

Identification of endophytic fungi of Papaver macrostomum, Chelidonium majus and Glaucium fimbrilligerum of Papaveraceae in Golestan province

Abstract
Endophytes are fungi that live within plants for at least a part of their life cycle without causing any visible manifestation of disease. Studies have shown that, nearly 300,000 plants species that exist on the earth, each individual plant is the host to one or more endophytes. Many endophytes have the potential to synthesize various bioactive metabolites. Many researches have proven that endophyte is new and potential source of novel natural products for exploitation in modern medicine, agriculture and industry. In order to identify the endophytic fungi from Papaver macrostomum, Chelidonium majus and Glaucium fimbrilligerum samples were taken from node of healthy plants from different locations in Gorgan, during the summer and autumn of 2017. Samples were surface sterilized with 70% ethanol (v/v) for 45 second, 1% sodium hypochlorite (v/v) for 30 second, and then washed three times with sterile distilled water. Samples were plated on potato dextrose agar culture medium and incubated at 25°C for 4 weeks. During this time, the fungi growing out of samples were isolated and purified by single spore and hyphal tip methods. Each isolate was identified by morphological characteristics such as conidia and conidiophore using appropriate identification keys. In order to identify non-spore isolates and verify the authenticity of the findings based on morphological characteristics, molecular identification and sequencing of ITS regions (ITS4 and ITS5) from the ribosomal DNA were performed and compared with GeneBank. In this study, 3 fungal species including Alternaria japonica, Alternaria arborescens and Lopadostoma fagi were identified as endophytic fungi of Papaveraceae. Alternaria species and a new genus reported for the first time form plants family of papaveraceae.
