چکیده
اندوفیت‏ها به عنوان موجوداتی با عفونت جزیی تعریف می‏ شوند که بافت‏های آلوده میزبانشان لااقل به ‏طور موقت بدون علائم بوده و کلنی میکروبی به‏ صورت داخلی است. در این تحقیق، به ‏منظور جداسازی و شناسایی قارچ‏های اندوفیت ریشه، طی سال 95-1394، از ریشه گیاهان سالم یکساله در مناطق مرکزی استان مازندران نمونه ‏برداری صورت گرفت. در آزمایشگاه، هر نمونه ریشه پس از سه مرحله شستشو و ضدعفونی سطحی با هیپوکلریت سدیم یک درصد به مدت یک ساعت، به قطعات 5 تا 10 میلی‏متری تقسیم و روی محیط PDA کشت و به مدت یک ماه در دمای 25 درجه سلسیوس و تاریکی مداوم، انکوبه شدند. خالص ‏سازی به روش نوک هیف صورت گرفت. خصوصیات فنوتیپی قارچ روی محیط کشت PDA در دمای 25 درجه سلسیوس بررسی شد. رشد پرگنه روی محیط کشت PDA کند، پر کردن پتری هشت سانتی‏متری در مدت یک ماه، ریسه ‏های هوایی بسیار کوتاه سفید رنگ در مرکز پرگنه، رنگ پرگنه از رو به ‏رو سیاه با مرکز سفید و در حالت وارونه به رنگ سیاه، حلقه سیاه حاشیه کلنی مملو از کلسیتوتسیوم‏های کروی به رنگ قهوه‏ ای تیره، به قطر 200-150 میکرومتر، آسک‏ها کروی، شفاف و حاوی هشت آسکوسپور، آسکوسپورها کروی، شفاف و 4-3 میکرومتر بودند. به منظور شناسایی مولکولی این جدایه، از استخراج DNA به روش CTAB و برای انجام واکنش زنجیره‏ای پلیمراز (PCR) از آغازگرهای ITS4 و ITS5 استفاده شد. توالی بدست آمده با استفاده از نرم‏ افزار بلاست در بانک ژن NCBI، با سایر توالی ‏های موجود در این بانک ژن، مورد مقایسه قرار گرفتند. این جدایه با هم‏پوشانی 98 درصد و با احتمال 99 درصد به عنوان گونه Pseudeurotium ovale شناسایی شد. در این پژوهش، براساس مطالعات ریخت ‏شناختی و توالی‏ یابی ناحیه ITS از DNA ریبوزومی، گونه Pseudeurotium ovale از ریشه کلزا به عنوان قارچ اندوفیت ریشه گیاهان یکساله جداسازی و شناسایی گردید و برای نخستین‏ بار این گونه از ایران گزارش می‏ شود. همچنین این نخستین گزارش این گونه از ریشه کلزا در دنیا می‏ باشد.

First report of Pseudeurotium ovale from Brassica napus roots in the north of Iran

Abstract
Endophytes are most commonly defined as those organisms which cause infections that are inconspicuous, the infected host tissues are at least transiently symptomless and the microbial colonization can be demonstrated to be internal. In this study, to separate and identify the root endophytic fungi, samples of annual plant roots were collected in the central regions of Mazandaran province during 2015-16. In the laboratory, each root sample was divided into 5-10 mm pieces after three stages of surface washing and disinfection, it was cultured on PDA medium and was incubated for a month at 25°C and in continuous darkness. Purification was carried out by using the hyphal tip method. Phenotyping characteristics of the isolate were investigated by growing on PDA at 25°C. The colony growth was slow on the PDA medium, filling eight centimeters of the petri dish in a month, very short white aerial mycelia in the center of the colony, the color of the colony was black with a white center and in the reverse, it was black, the black ring of the colonial margin was filled with a dark brown spherical cleistothecium, ranging in diameter from 150-200 μm, spherical, transparent, the asci were spherical and translucent with ellipsoidal and hyaline eight ascospores, 3-4 μm. In order to molecular identification of this isolate, the DNA was extracted by the CTAB method, and ITS4,5 primers were used to perform polymerase chain reaction (PCR). The sequences were compared with the other sequences in the NCBI, by using the Blasts software in this gene bank. This isolate was identified by 98% query coverage and 99% probably as Pseudeurotium ovale species. In this research, on the basis of morphological studies and sequencing of the ITS region from the rDNA, Pseudeurotium ovale from the root of canola was identified as the endophytic fungus of the annual plants and for the first time, this species is reported from Iran. It is also the first report of this species of canola roots in the world.
