تاثیر نماتودهای بیمارگر حشرات Steinernema carpocapsae و S. feltiae روی کرم برگخوار چغندر Spodoptera exigua

چکیده
نماتود‌های بیمارگر حشرات متعلق به خانواده‌ Steinernematidae به‌عنوان پاتوژن‌های اجباری و کشنده‌ی حشرات قادر به کنترل طیف وسیعی از آفات با اهمیت اقتصادی بالا هستند. این نماتودها به‌دلیل داشتن ویژگی‌های مانند زهرآگینی بالا، قابلیت تولید انبوه، میزبان‌یابی سریع و سازگار‌ی با اکثر تجهیزات رایج سم‌پاشی یکی از موفق‌ترین عوامل کنترل زیستی می‌باشند. از سوی دیگر بررسی‌های صورت گرفته نشان داده که گونه‌ها و جدایه‌های مختلف کارایی‌های متفاوتی روی آفات خاص و در شرایط خاص نشان می‌دهند. کرم برگخوار چغندر یک آفت پلی‌فاژ می‌باشد که عموما کنترل آن متکی بر استفاده از سموم شیمیایی می‌باشد و لذا جهت جلوگیری از القای مقاومت، از بین رفتن دشمنان طبیعی و آلودگی زیست محیط لازم است راهکارهای سالم جایگزین یا مکمل توسعه یابد. استفاده از عوامل کنترل بیولوژیک می‌تواند راه‌کار بسیار موفقی در رابطه با کنترل این آفت باشد. با هدف بررسی کارایی جدایه‌های بومی در کنترل این آفت، از لارو 24 ساعته جهت زیست‌سنجی استفاده گردید. لاروهای آلوده‌کننده نماتودها با غلظت مشخص (6،12،25،50،100،200،300 نماتود به ازای یک لارو) به روی کاغذ صافی اسپری، لارو‌ها روی آن رهاسازی و مرگ و میر به مدت 72 ساعت روزانه ثبت شد. در هر دو گونه با افزایش غلظت نماتود مرگ و میر افزایش یافت، در گونه‌ی S. carpocapsae (F6, 21 = 59.4; P < 0.05) در عرض 24 ساعت، بیشترین (100%) و کمترین (10%) مرگ و میر به‌ترتیب در غلظت‌های 200 و 7 IJ/L و در گونه‌‌ی S. feltiae (F6, 21 = 7.9; P < 0.05) در غلظت‌های 100 و 7 IJ/L (به ترتیب 60% و 10% مرگ و میر) ثبت گردید. با افزایش غلظت‌ بالاتر از 200 IJ/L در گونه‌ی S. carpocapsae و بالاتر از 100 IJs/L در گونه‌‌ی S. feltiae تغییری در میزان مرگ و میر پس از 24 ساعت ثبت نگردید. با افزایش زمان آلوده‌سازی در هر دو گونه و با گذر زمان میزان مرگ و میر افزایش یافت، در گونه S. carpocapsae پس از 72 ساعت بیشترین (100%) و کمترین (60%) میزان مرگ و میر به ترتیب در غلظت‌-های 200 و 7 IJ/L و در گونه S. feltiae بیشترین (100%) و کمترین (70%) میزان مرگ و میر به ترتیب در غلظت‌‌های 25 و 7 IJ/L ثبت گردید. کارایی متفاوت این دو گونه نماتود می‌تواند نه تنها ناشی از تفاوت در گونه آنها، بلکه ناشی از تفاوت در بیماریزایی باکتری‌های همزیست و استراتژی‌های میزبان‌یابی نماتودها باشد. در مجموع نتایج نشان داد گونه‌‌ی S. feltiae در شرایط آزمایشگاهی کارایی کنترلی بیشتری روی لاروهای سن اول آفت دارد.

Efficacy of two entomopathogenic nematodes, Steinernema carpocapsae and S. feltiae against Beet Armyworm, Spodoptera exigua

Abstract
Entomopathogenic nematodes (EPN) from the family Steinernematidae are obligate insect pathogen, capable of infecting and killing a number of economically important insect pests. EPNs can be considered useful biological control agents due to high virulence, efficient host strategy, ease of mass production and safety towards non-target organisms. Different species and isolates of EPNs show different levels of efficacy against pests. Beet armyworm, Spodoptera exigua, is one of the most important cosmopolitan and polyphagous pest that cause serious damage. This pest is mostly controlled using synthetic insecticides then effective alternatives to synthetic insecticides such as biocontrol method need to be developed to reduce the pressure for insecticide resistance development and to reduce negative effects of the insecticides on natural enemies of pests and other non-target organism including humans. With the aim of evaluate the efficacy of native isolates of Steinernema carpocapsae and S. feltiae, 24 hours beet armyworm larvae susceptibility were studied using filter paper assays during 72 hours of post infection. EPNs efficacy were studied using seven treatments including 6, 12, 50, 100, 200 and 300 across four replications. Control treatment received only Ringer's solution. Beet armyworm mortality under treatments of both EPNs species were significantly affected. Unlike S. feltiae (F6, 21 = 7.9; P < 0.05) in which the mortality reached to its highest rate (60%) at 100 IJs/L concentration after 24 hours of post infection, larvae mortality exposed to S. carpocapsae (F6, 21 = ; P < 0.05) increased up to 200 IJ/L (100%) then plateau, at the same period, lowest rate of mortality of both EPNs were recorded at 10% IJs/L. By increasing the post infection period, mortality increased, after 72 hours and under S carpocapsae treatment, highest (%100) and lowest (%60) mortality were recorded at 200 IJs/L and 7 IJs/L respectively, these values for S. feltiae were 100 and 70 recorded at 25 IJs/L and 7 IJs/L respectively. In our results S. feltiae appeared as a more efficient EPN against 24 hours larvae. These differences in virulence can be attributed not only to EPNs inter-specific differences but also to their symbiotic bacteria or host strategies.
