شناسایی قارچ‌های همراه با نماتد سیستی غلات (Heterodera filipjevi) در استان آذربایجان شرقی

چکیده
گونه Heterodera filipjevi از منظر پراکنش و اقتصادی یکی از مهم‌ترین گونه-های نماتدهای سیستی غلات در جهان است. نماتدهای سیستی غلات از استان‌های مختلف کشور به ویژه استان آذربایجان شرقی گزارش شده‌اند. کنترل زیستی به عنوان یک راه‌کار مدیریتی همراه با دیگر روش‌ها برای کنترل نماتدهای انگل گیاهی از جمله این نماتدها مورد توجه قرار گرفته است. از میان دشمنان طبیعی مختلفی که روی نماتدهای انگل گیاهی اثر می گذارند، درصد بیشتری به قارچ های آنتاگونیست تعلق دارد. تحقیق حاضر با هدف شناسایی قارچ‌های همراه نماتد سیستی غلات، H. filipjevi، در استان آذربایجان شرقی انجام شد. برای این منظور، از خاک‌های آلوده به H. filipjevi در مناطق عمده کشت گندم استان شامل مراغه، هشترود و سراب نمونه‌برداری صورت گرفت. سیست‌ها با استفاده از روش فنویک از نمونه‌های خاک جداسازی شده و پس از ضدعفونی سطحی با هیپوکلریت سدیم یک درصد، روی محیط غذایی PDA و WA کشت گردیدند. پتری‌ها به مدت ده روز در دمای 25 درجه سانتی‌گراد نگهداری و جدایه‌های قارچی رشد کرده با روش تک اسپور خالص‌سازی شدند. شناسایی جدایه‌های قارچی با استفاده از صفات ریخت‌شناختی و داده‌های توالی نوکلئوتیدی ناحیه ITS-rDNA انجام شد. نتایج نشان داد که تنوع گونه‌ای قابل توجهی در گونه‌های قارچی کلونیزه کننده H. filipjevi وجود دارد. در این تحقیق قارچ‌های Alternaria chlamydosporigena، Microdochium bolleyi، Chaetomium globosum، Oidiodendron cerealis، Exophiala equina، Periconia macrospinosa، Trichocladium opacum، Auxarthron alboluteum و Cadophora sp. شناسایی شدند. از این بین، Microdochium bolleyi، Oidiodendron cerealis، Exophiala equina، Periconia macrospinosa، Trichocladium opacum و Auxarthron alboluteum گزارش‌های جدیدی برای فلور قارچی ایران هستند. همچنین، سیست H. filipjevi به عنوان میزبان جدید برای تمام گونه‌های قارچی فوق به‌جز گونه C. globosum گزارش می-گردد.

Characterization of the fungi associated with cereal cyst nematode (Heterodera filipjevi) in East Azarbaijan province

Abstract
Heterodera filipjevi is one of the most important species of cyst nematodes in the world in terms of distribution and economics. Cyst nematodes of cereals have been reported from different provinces of the country, especially East Azarbaijan province. Biological control along with other methods has been considered as a management approch for control of parasite nematodes, including these nematodes. Among the various natural enemies that affect plant nematodes, a great percentage includes the antagonistic fungi. The aim of present study was to identify fungi associated with cyst nematode of cereals, H. filipjevi, in East Azarbaijan province. For this purpose, soils infested with H. filipjevi were sampled from main areas of wheat growing in the province including Maragheh, Hashtrood and Sarab regions. The cysts were extracted from soil samples using the Fenwick method, surface sterilized with 1% sodium hypochlorite and plated on PDA and WA culture media. Plates were incubated at 25 °C for 10 days and pure cultures were established from fungal colonies using single spore method. The results obtained in the present study revealed that a rich species diversity exist in fungi colonizing H. filipjevi. In this study Alternaria chlamydosporigena, Microdochium bolleyi, Chaetomium globosum, Oidiodendron cerealis, Exophiala equina, Periconia macrospinosa, Trichocladium opacum, Auxarthron alboluteum and Cadophora sp. were identified. Of these, Microdochium bolleyi, Oidiodendron cerealis, Exophiala equina, Periconia macrospinosa, Trichocladium opacum and Auxarthron alboluteum are new reports for mycobiota of Iran. Furthermore, cyst of H. filipjevi is reported as a new host for all the above mentioned fungi except C. globosum species.
